Two-time defending champion Barbora Krejčíková and her partner, Rajeev Ram, defeated Samantha Stosur and Matthew Ebden in the final, 6–1, 6–4, to win the mixed doubles tennis title at the 2021 Australian Open. This was the second major mixed doubles title Krejčíková and Ram won together as a team after they won their first at the 2019 Australian Open. The pair saved a match point in their second round match against Ena Shibahara and Ben McLachlan en route to the title.[1]

Krejčíková had won the preceding year's tournament with Nikola Mektić,[2] but they had chosen to participate with different partners in 2021. Mektić played alongside Barbora Strýcová, but lost in the first round to Hayley Carter and Sander Gillé.

After not being held at the 2020 US Open and 2020 French Open, this year's tournament marked the return of Mixed Doubles to a Grand Slam amid the COVID-19 pandemic.[3]
